To add to that, if you need to manually approve someone, you can do so by visiting the profile, clicking Edit, then "Activate user" in the modal.
Though in practice this shouldn't be needed if your outbound email works.
If a user took too long to click the validation link, they can login and click on the banner to request a new email, or use the password reset feature which will also enable their account once a new password is set.
